It is broadly trusted that customary pharmaceutical here and there called herbalism is the most antiquated strategy for curing illnesses (Evans, 1989). It has been realized that plants are the first and final genuine drugs ever utilized by man. Be that as it may, in Nigeria, up to this point, the acts of the utilization of herbs have been kept mystery and covered in feared supernatural chants, ceremonies and penances. It is as of now clear that the quality of the plants and its parts does not depend on upon such appear. The use of plants for remedial aims is a vital mostly of life and custom in Africa. According to Ekhaise (2002), around 80% of the populace depends specifically on the conventional medication for essential medicinal services.

Medicinal plants are of awesome significance to the soundness of people and groups; it has been utilized by numerous individuals to cure their sicknesses. The therapeutic estimations of these plants lie in some compound substances that create unequivocal physiological changes in human body. The most critical of these bioactive constituents of plants are alkaloids, tannins, flavonoids, steroids, and phenolic mixes (Edeoga et al, 2005). A considerable lot of these indigenous restorative plants are utilized as flavors and sustenance plants. Therapeutic plants are of incredible significance to the strength of people and groups (Edeoga et al., 2005). A considerable lot of these indigenous therapeutic plants are utilized as flavors and nourishment plants. They are some of the time added to nourishments implied for pregnant and nursing moms for restorative purposes.

Literature review

The therapeutic estimation of plants have expected a more essential measurement in the previous couple of decades owing to a great extent to the disclosure that concentrates from plants contain minerals and essential metabolites as well as a differing cluster of optional metabolites with antidiabetic potential (Denni and sadiq, 2000). In any case, the act of home grown drug in the creating parts of the globe needed experimental premise or models, in this way constraining their conceivable advantages. As stated by Dicko (2005), to cure this and guarantee their maximal use and viability the quality and consistency of these home grown solutions ought to be found out and looked after.

Guineense schum and thom, usually called West Africa dark pepper (Piperaceae) is a herbaceous climber normally found in the African tropical timberland zones, with over more than 700 species all through the tropical and subtropical districts of the world11. In Nigeria, it is known with various vernacular names like uziza and iyere among the Igos and Yoruba tribes, individually. P. guineense is fundamentally the same to Piper nigrum and has culinary, restorative, corrective and insecticidal applications. The natural products or berries and leaves are generally sold in Southern Nigerian markets as sauces and for sustenance enhancing. The dietary sythesis of P. guineense leaf was given as 175 mg/100g DM ascorbic corrosive, 4.2 mg/100g DM starch, 3.8 mg/100g DM protein, 76% dampness and 2.9 mg/100gDM fiber. The leafy foods are broadly utilized as a part of Nigerian customary medication as a solution for ailment, bronchitis, hack, stomach issue, intestinal maladies, gonorrhea and ripeness upgrade in ladies. The fluid concentrate of the ground seeds and leaves is controlled to nursing moms, for the standardization of the womb after conveyance and as flavors for soup (pepper soup) to patients having fever. The seed and leaf concentrates of this plant had been accounted for to have antimicrobial exercises, hepatoprotective impact, insecticidal impact, antifertility impact and cholinergic movement in uterine muscle reaction (Ogu et al, 2012). They contain a lot of beta-carophyllene, which is being researched as a calming specialist. Their utilizations in ethnomedicine incorporate going about as stimulants, germicide, carminatives, diuretics, laxatives, anticonvulsant, antihelmitic, and narcotics for treatment of loose bowels, jungle fever, ailment, asthma, catarrh and bronchitis (Ndukwu and Ben, 2003). Leaves of guinnense are utilized for respiratory diseases and for female fruitlessness while its natural products are utilized as a sexual enhancer (Mbongue et al, 2005). The leaves of iyere additionally serve as home grown cure for hypertension, looseness of the bowels, low sperm tally and menstrual issue (Prof. olowokudejo, 2004). The sabanut is a perfect sub-tropical sustenance tree that is dry spell, infection, and nourishment tolerant. It is a strong plant and will adjust exceptionally well to various conditions. The pachira needs a lot of daylight however it is essential to keep away from direct daylight amid the mid year months as the leaves may get sunburned. In Hawaii, it is at times known as Malabar Chestnut. In Central American, it is known as the procurement tree. Saba seeds are palatable in the wake of being cooked and have an aftertaste like chestnut. At the surrounding temperature of the tropics( the separated fat) has the consistency of petroleum jam, is white in shading and consumable with a wonderful smell faintly fragrant of licorice. The fat (which has a modern potential) can be utilized as a part of making cleanser, yet would be better used on the off chance that it were refined into a consumable item (Edwin, 1977).  Alongside basic auxiliary metabolites like; alkaloid, flavonoids, coumarins and terpenes, various substances, as; tannins, gallic corrosive, quercetin, phenols, phyto-sterols, alcohols, and so forth have been accounted for in mistletoe plants through utilizing thin layer chromatography (TLC). The identification of dynamic standards in therapeutic plants assumes a vital part in both subjective and quantitative phytochemical examination of rough plant separates (Laurence et al., 2003). TLC is a quick and prudent methodology for the determination of the primary dynamic standards of therapeutic plants e.g., alkaloids, cardiovascular glycosides, coumarins, flavonoids, saponins, tannins, and so on. TLC is likewise utilized for fractionation of the concentrate got by extraction methodology by utilizing diverse dissolvable structures.

PHYTOCHEMICALS

According to Harborne (1973), Phytochemicals are characterized as bioactive non-supplement plant mixes in organic products, vegetables, grains, and other plant nourishments that have been connected to diminishing the danger of major incessant illnesses. The therapeutic estimations of these plants lie in some compound substances that deliver clear physiological changes in human body. The most critical of these bioactive constituents of plants are alkaloids, tannins, flavonoids, ketonic mixes, terpenoids, steroids, and phenolic mixes (Edeoga et al, 2005). By and large "phyto" from phytochemical is a greek word which implies plant. Along these lines phytochemicals are plant chemicals.

WHY PHYTOCHEMICALS SHOULD BE IN OUR DIET

At the point when devoured in the eating regimen, there is an expanding assemblage of proof to show that phytochemicals may diminish the danger of age-related perpetual illnesses, for example, coronary illness, diabetes, hypertension and certain sorts of growth. In spite of the fact that their nonattendance from the eating regimen won't not bring about inadequacy side effects, for example, those found with vitamins and minerals, they are thought to be critical for wellbeing and prosperity all through life, particularly in adulthood and in the elderly. Phytochemicals are thought to go about as synergistic operators, permitting supplements to be utilized all the more productively by the body. Collaboration is the cooperating of two things (sustenance segments, for instance) to create an impact more noteworthy than the aggregate of their individual impacts (Edeoga et al, 2005).

FOOD SOURCES OF PHYTOCHEMICALS

Phytochemicals are found in all plant items. It is exhorted that we expend a wide assortment of products of the soil with a specific end goal to increase most extreme advantage from the supplements and phytochemicals they contain. Ideally, admission of phytochemicals ought to be from dietary sources as opposed to from supplements or pills. These can just give a couple of the a huge number of phytochemicals accessible to us and are in this manner less compelling than a serving of fruits and vegetables.

3   BIOACTIVE CONSTITUENTS OF PLANTS

1.3.3.1 PHENOLICS

The phenolic acids are plant metabolites generally spread all through the plant kingdom. Late enthusiasm for phenolic acids originates from their potential defensive part, through ingestion of products of the soil, against oxidative harm maladies (coronary illness, stroke, and growths). Phenolic mixes are fundamental for the development and propagation of plants, and are created as a reaction for shielding harmed plants against pathogens. Phenolic compound are: Flavonoids, cuomarins, tannins, steroids (Hertog et al, 1993).

1.3.3.2   FLAVONOID

Flavonoids are gathering of phenolic mixes having cancer prevention agent action and have been accounted for to have antiviral, subterranean insect iallergic, mitigating and cell reinforcement exercises. Flavonoids are polyphenolics aggravates that are pervasive in nature and are ordered, by structure, into flavonols, flavones, flavanones, isoflavones, catechins, anthocyanidins and chalcones. More than 4,000 Flavonoids have been recognized, a large portion of which happen in organic products, vegetables and refreshments. The limit of Flavonoids to go about as cancer prevention agents relies on their atomic structure. The position of hydroxyl gatherings and other element in the synthetic structure of Flavonoids are imperative for their cell reinforcement exercises (Hertog et al, 1993).

SAPONINS

Saponins are glycosides with a particular frothing trademark. They are found in numerous plants, yet get their name from the soapwort plant (saponaria), the foundation of which was utilized truly as a cleanser. They comprise of polycyclic aglycone that is either a choline steroid or triterpenoid appended through C3 and an ether bond to a sugar side chain. The aglycone is alluded to as the sapogenin and steroid Saponin are called saraponins.

The capacity of Saponin to froth is brought about by the mix of the nonpolar sapogenin and the water solvent side chain. Saponins are severe and diminish the acceptability of animals sustains. Saponins are broadly being explored for cholesterol control. The blood cholesterol bringing down properties of dietary saponins are specifically compelling in human sustenance. Saponins have been observed to been to be valuable in the treatment for hyperchlosterolemia.

TANNINS

Tannins has astringent properties, hurries the mending of wounds and excited mucous film. The nearness of tannins in the leaves of Spondias Mombin can bolster its solid use for mending of wounds, varicose ulcers, hemorrhoids, ice nibble and blaze in home grown prescription (Agnaniet, 2005). Tannins are actually happening complex polyphenolics found in numerous plants, especially those with a woody propensity. Their fundamental capacity in nature is by all accounts one of assurance; creatures are dissuaded from eating plants high in tannins in view of the intense, astringent taste. In spite of the fact that generally concentrated on in horticultural circles subsequent to 1960's, tannins have been the subject of almost no restorative exploration. The human studies that have been done for the most part concern the impacts of tannins with respect to tumor and coronary illness.

STEROIDS

Steroid is a sort of natural exacerbate that contains a trademark game plan of four cycloalkane rings that are joined to each other. Case of steroids incorporates the dietary fat cholesterol, sex hormones estradiol and testosterone. Steroids fluctuate by the utilitarian gathering joined to this four ring center and by the oxidation condition of the ring.

ALKALOIDS

Alkaloids are a gathering of normally occuring concoction exacerbates that contain for the most part essential nitrogen molecules. This gathering likewise incorporates some related mixes with nonpartisan and even pitifully acidic properties. Additionally some engineered mixes of comparable structure are ascribed to alkaloids. Notwithstanding carbon, hydrogen and nitrogen, alkaloids may likewise contain oxygen, sulfur and all the more once in a while different components, for example, chlorine, bromine, and phosphorus. Alkaloids are delivered by a huge assortment of living beings, including microscopic organisms, growths, plants, and creatures, and are a piece of the gathering of characteristic items (likewise called optional metabolites). Illustrations are the nearby soporific and stimulant cocaine; the hallucinogenic psilocin; the stimulant caffeine; nicotine; the pain relieving morphine; the antibacterial berberine; the anticancer compound vincristine; the antihypertension specialist reserpine; the cholinomimeric galatamine; the spasmolysis operator atropine; the vasodilator vincamine; the counter arhythmia compound quinidine; the counter asthma restorative ephedrine; and the antimalarial drug quinine. In spite of the fact that alkaloids follow up on a differing qualities of metabolic frameworks in people and different creatures, they consistently summon a biting taste (Hertog et al, 1993)

KETONIC COMPOUNDS

The word ketone gets its name from Aketon, an old German word Acetone. As indicated by the guidelines of IUPAC classification, ketones are named by changing the postfix – ane of the guardian alkane to – one. For the most imperative ketones, notwithstanding, customary nonsystematic names are still by and large utilized, for instance acetone and benzophenone. Ketone mixes have critical physiological properties. They are found in a few sugars and in mixes for therapeutic use, including normal and manufactured steroid hormones. The most essential ketone is CH3)2CO, a fluid with a sweetish scent, which makes it simple to evacuate by dissipation when didn't really needed. It is a standout amongst the most vital mechanical solvents being utilized as a part of such items as paints, varnishes, pitches and coatings or nail-shine removers (Hertog et al, 1993)

ANTHRAQUINONES

Anthraquinones actually exist in some plants (aloe),fungi, lichen, and bugs where it serves as an essential skeleton for their colors. Normal anthraquinones subsidiaries have a tendency to have purgative impacts (A.I vogel, 2001). Anthraquinones is utilized as a part of generation of wood mash in mash and paper industry. Another utilization is as a repellant on seeds. Anthraquinones (9,10-dioxoanthracene) is a sweet-smelling natural compound. It is a subordinate of anthracene, it resembles yellow or light dim to dim green strong crystalline powder. Its chemical formular is C14H8O2.

CARDIAC GLYCOSIDES

Cardiovascular Glycosides has a place with the gathering of cardioactive medications. Be that as it may, the restorative utilization of medications for the treatment of the fizzling heart is affirmed to a little gathering of plant glycosides which acts straightforwardly on the heart muscle. A significant number of plants scattered all through the plant kingdom contain C23 and C24 steroidal glycosides which apply on the fizzling heart a moderating and reinforcing impact.

Plants heart glycosides seem, by all accounts, to be limited to the angiosperms. The sugar moieties, connected to the anglycone by C-2, β-linkage are made out of up to three sugar units which may incorporate glucose or mannose together with other deoxy – sugar whose normal event is known just in relationship with cardiovascular glycosides. The pharmacological adequacy of cardio-dynamic glycosides is reliant on both aglycones and the sugar connections, the inalienable action lives in the aglycones, however the sugar render the mixes more dissolvable and expansion the power of myocardiac withdrawal. The heart capturing properties of these glycosides likewise render them best as bolt toxic substances.

2.2.1 SAMPLE COLLECTION AND IDENTIFICATION

The fresh leaves of iyere, and sabanut leaves were collected from Federal College of Agriculture, Akure (FECA) and were taken to the department of Crop, Soil, and Pest Management, Federal University of Technology, Akure (FUTA) for identification. The plant samples were dried for two weeks.

2.2.2   SAMPLE PREPARATION

The plant samples were air-dried for two weeks and ground into powdery form with a blender. The purpose of the grinding is to increase the surface area of the samples for easy extraction. The powdered samples were weighed on an analytical balance for the various phytochemical tests.

2.3 PHYTOCHEMICAL SCREENING

2.3.1   TEST FOR TANNINS

About 0.5 g of the dried powdered sample was boiled in 20 ml of water in a test tube and after that separated. A couple drops of 0.1% ferric chloride was included and watched for tanish green or a blue-dark shading.

2.3.2 TEST FOR SAPONIN

Around 2 g of the powdered example was bubbled in 20 ml of refined water in a water shower and separated. 10ml of the filtrate was blended with 5 ml of distilled water and shaken energetically for a stable relentless foam. The foaming was blended with 3 drops of olive oil and shaken vivaciously, then watched for the development of emulsion.

2.3.3 TEST FOR FLAVONOIDS

5 ml of weaken NH3(aq) solution was blended to an aliquot part of the separated segment of both plant extricates from that point took after by expansion of concentrated H2SO4. A yellow colouration found in each concentrate demonstrated the proximity of flavonoids. The yellow colouration vanished on standing.

Additionally a segment of the powdered plant test was for every situation warmed with 10 ml of ethyl acetic acid derivation over a steam shower for 3 min. The blend was sifted and 4 ml of the filtrate was shaken with 1 ml of the ammonium solution. A yellow colouration was watched demonstrating a positive test for flavonoids.

2.3.4 TEST FOR CARDIAC GLYCOSIDES (KELLER-KILLANI TEST)

5ml of each plant concentrates was treated with a mixture of 2 ml of glacial acetic acid containing one drop of FeCl3 solution. This was underlayed with 1 ml of concentrated H2SO4. A brown ring of the interface demonstrates a deoxysugar normal for cardenolides. A greenish ring was watched showing the nearness of heart glycosides.

2.4  THIN LAYER CHROMATOGRAPHY (TLC) METHOD

2g of each powdered samples was absorbed with 5 ml of methanol and were left to remain for 7 days, supernatant layer was utilized for the TLC examination. The TLC plates (10 by 10cm glass plates) were made by blending the adsorbent (silica gel) containing little measure of inactive folio calcium sulfate (gypsum) with double the measure of water. The blend was spread as slurry on the already cleaned glass miniaturized scale plates utilizing the TLC spreader. The plates were set and afterwards dried and initiated at 110°C in a stove for 30 min. The concentrates were drawn with narrow tubes and connected as spots on a stationary stage (silica-gel covered plate) around 1 cm from the base. The plate was then dunked into a reasonable dissolvable framework (versatile stage) and put in a very much secured tank.

The following solvent systems were used:

Solvent system I: Ethyl acetate: Toluene 8:4

Solvent system II: Ethyl acetate: Toluene: Acetic acid 8:4:1

Chromatographic tank was saturated with mobile phase at room temperature for 5 min prior to development.

At the end of the chromatographic development which lasted for about 20 min, the plate was removed from the chromatographic tank and then allowed to dry for few minutes. Lines were drawn 1cm from both ends of the plates and the separated spots were visualized under daylight and after spraying with chromogenic reagents. Distances between the spots were measured and the retention factor (Rf) values were recorded, using the following formular:

Rf value = (Distance moved by the compound)

(Distance moved by the solvent front)

The presence of secondary metabolites in the leaf extracts were

qualitatively determined by TLC using;

1. Dragendorff’s reagent and HClO4 spray reagent for the

presence of alkaloids.

2. Dragendorff’s reagent and Fecl3 spray reagent for the

presence of alkaloids.

3. 5% methanolic KOH for the presence of anthraquinones.

4. 2, 4 – dinitrophenyl hydrazine for the presence of ketonic compounds.
